    Atherosclerosis regression and TP receptor inhibition: effect of S18886 on plaque size and composition—a magnetic resonance imaging study

    Aims Endothelial dysfunction, platelet hyperactivity, and inflammation play a crucial role in atherogenesis. A growing body of evidence suggests that inhibition of the thromboxane A2 (TxA2 or TP) receptor may improve endothelial function and reduce the inflammatory component of atherosclerosis in addition to its demonstrated antiplatelet activity. Consequently, we sought to assess the effect of a novel TP receptor antagonist S18886, on atherosclerotic lesion progression and composition by serial non-invasive magnetic resonance imaging (MRI). Methods and results S18886 was compared with control in an experimental model of established aortic atherosclerosis in New Zealand White rabbits (n=10). The animals underwent MRI of the abdominal aorta at the time of randomization and at the end of treatment. Subsequently, animals were euthanized and specimens were stained for histopathology and immunohistochemistry with anti-α-actin antibodies for vascular smooth muscle cells (VSMC), anti-RAM-11 for macrophages, anti-caspase-3 for apoptotic cells, anti-MMP-1 for metalloproteinases, and anti-endothelin-1 (ET-1) as a marker of endothelial dysfunction. MRI analysis revealed a significant reduction in total vessel area (TVA) and vessel wall area (VWA) in the S18886 group (P<0.05). Immunostaining analysis showed a significant decrease in RAM-11, caspase-3, MMP-1, ET-1 and an increase in α-actin in the treated group (P<0.05 vs. control). Conclusion Inhibition of the TP receptor by S18886 causes a regression of advanced atherosclerotic plaques. In addition, the reduction in the markers for macrophages, apoptotic cells, metalloproteinases, and endothelin-1 and the increase in VSMC, suggests that S18886 may not only halt the progression of atherosclerosis, but also transform lesions towards a more stable phenotype. The possibility of combining antithrombotic and antiatherosclerotic activity by means of the administration of TP inhibitors deserves further investigation in a clinical settin

    Recent Advances in the Processing and Rendering Algorithms for Computer-Generated Holography

    Digital holography represents a novel media which promises to revolutionize the way the users interacts with content. This paper presents an in-depth review of the state-of-the-art algorithms for advanced processing and rendering of computer-generated holography. Open-access holographic data are selected and characterized as references for the experimental analysis. The design of a tool for digital hologram rendering and quality evaluation is presented and implemented as an open-source reference software, with the aim to encourage the approach to the holography research area, and simplify the rendering and quality evaluation tasks. Exploration studies focused on the reproducibility of the results are reported, showing a practical application of the proposed architecture for standardization activities. A final discussion on the results obtained is reported, also highlighting the future developments of the reconstruction software that is made publicly available with this work

    Structural and Functional Characterization of a New Double Variant Haemoglobin (HbG-Philadelphia/Duarte α268Asn→Lysβ262Ala→Pro)

    We report the first case of cosegregation of two haemoglobins (Hbs): HbG-Philadelphia [α68(E17)Asn → Lys] and HbDuarte [β62(E6)Ala → Pro]. The proband is a young patient heterozygous also for β°-thalassaemia. We detected exclusively two haemoglobin variants: HbDuarte and HbG-Philadelphia/Duarte. Functional study of the new double variant HbG-Philadelphia/Duarte exhibited an increase in oxygen affinity, with a slight decrease of cooperativity and Bohr effect. This functional behaviour is attributed to β62Ala → Pro instead of α68Asn → Lys substitution. Indeed, HbG-Philadelphia isolated in our laboratory from blood cells donor carrier for this variant is not affected by any functional modification, whereas purified Hb Duarte showed functional properties very similar to the double variant. NMR and MD simulation studies confirmed that the presence of Pro instead of Ala at the β62 position produces displacement of the E helix and modifications of the tertiary structure. The substitution α68(E17)Asn → Lys does not cause significant structural and dynamical modifications of the protein. A possible structure-based rational of substitution effects is suggested

    Rasagiline withdrawal Syndrome in Parkinson’s Disease

    Parkinson’s disease (PD) patients using dopamine agonists can develop withdrawal symptoms, referred to as dopamine agonist withdrawal syndrome (DAWS), under dose tapering or discontinuation of these drugs. DAWS includes a severe stereotypical cluster of psychiatric and psychological symptoms encompassing severe mood and anxiety disturbances, autonomic symptoms, as well as generalized pain and drug cravings. However, symptoms of withdrawal of dopamine replacement therapies (DRT) are not simply limited to dopamine agonists tapering, as observed in PD patients on deep brain stimulation after dopaminergic drugs withdrawal related to surgery. To date, no DRT-related withdrawal syndrome has been described in PD patients who discontinue rasagiline, an irreversible inhibitor of monoamine oxidase-B (MAO-B). Here we report three PD patients who developed a severe withdrawal syndrome after rasagiline suspension. The syndrome was mainly characterized by prominent psychiatric disorders (depression, anxiety with panic attacks, dysphoria, and agitation) associated with fatigue, generalized pain, and autonomic manifestations (closely resembling symptoms of DAWS). In our opinion, this report suggests the importance of closely monitoring PD patients undergoing rasagiline suspension for withdrawal symptoms and provides interesting points of reflection on the role of rasagiline and other MAO-B inhibitors in mood disorders

    Association of kidney disease measures with risk of renal function worsening in patients with type 1 diabetes

    Background: Albuminuria has been classically considered a marker of kidney damage progression in diabetic patients and it is routinely assessed to monitor kidney function. However, the role of a mild GFR reduction on the development of stage 653 CKD has been less explored in type 1 diabetes mellitus (T1DM) patients. Aim of the present study was to evaluate the prognostic role of kidney disease measures, namely albuminuria and reduced GFR, on the development of stage 653 CKD in a large cohort of patients affected by T1DM. Methods: A total of 4284 patients affected by T1DM followed-up at 76 diabetes centers participating to the Italian Association of Clinical Diabetologists (Associazione Medici Diabetologi, AMD) initiative constitutes the study population. Urinary albumin excretion (ACR) and estimated GFR (eGFR) were retrieved and analyzed. The incidence of stage 653 CKD (eGFR &lt; 60 mL/min/1.73 m2) or eGFR reduction &gt; 30% from baseline was evaluated. Results: The mean estimated GFR was 98 \ub1 17 mL/min/1.73m2 and the proportion of patients with albuminuria was 15.3% (n = 654) at baseline. About 8% (n = 337) of patients developed one of the two renal endpoints during the 4-year follow-up period. Age, albuminuria (micro or macro) and baseline eGFR &lt; 90 ml/min/m2 were independent risk factors for stage 653 CKD and renal function worsening. When compared to patients with eGFR &gt; 90 ml/min/1.73m2 and normoalbuminuria, those with albuminuria at baseline had a 1.69 greater risk of reaching stage 3 CKD, while patients with mild eGFR reduction (i.e. eGFR between 90 and 60 mL/min/1.73 m2) show a 3.81 greater risk that rose to 8.24 for those patients with albuminuria and mild eGFR reduction at baseline. Conclusions: Albuminuria and eGFR reduction represent independent risk factors for incident stage 653 CKD in T1DM patients. The simultaneous occurrence of reduced eGFR and albuminuria have a synergistic effect on renal function worsening

    Design di interfacce e usabilità del Web in ambiente mobile: la Programmazione della Cineteca di Bologna

    Il presente progetto è stato svolto in collaborazione con la Cineteca di Bologna e verte sulla riprogettazione dell'odierna interfaccia Web, in funzione della sua visualizzazione mobile non considerata nella progettazione del 2008, in particolare nella sua parte che concerne la programmazione degli spettacoli in ambito cinematografico. Raccolti i suggerimenti degli utenti e approfondite le richieste della Cineteca, affrontando con i responsabili IT della Fondazione le criticità emerse, la ristrutturazione delle sezioni presenti e l'aggiunta di nuove funzionalità, si è impostato il lavoro come segue. In primis, si è eseguita una ricerca, avvalendosi di fonti autorevoli nella letteratura di settore, con un focus incentrato sull’Usabilità. Questa ha portato a presentare una panoramica di tale tematica, per concentrarsi poi sulle peculiarità dei dispositivi mobili, toccando vari aspetti: da un approfondimento dalle caratteristiche dell’uso fino una collocazione anche storica alla realtà mobile. Successivamente, si è proseguito analizzando nel dettaglio tutti i vari elementi da includere, sia dal punto di vista concettuale che posizionale, in riferimento a varie soluzioni proposte dalla letteratura di settore. Ciò preferendo i modelli principalmente utilizzati, dunque maggiormente familiari agli utenti, e i metodi normalizzati per la risoluzione di singole problematiche frequenti nella presentazione delle informazioni. Il risultato è un progetto di interfaccia più vicina all'esperienza quotidiana dell'utente, concretizzatosi con la realizzazione di un prototipo per mezzo di applicativi che simulano un device e mostrano anteprime grafiche dell'interfaccia medesima, nella fattispecie della natura e della collocazione dei singoli componenti sullo schermo. Il lavoro, pertanto, si pone l’obiettivo di rispondere a comuni e debite aspettative dell'utenza in fatto di comodità, efficienza e immediatezza di consultazione della Programmazione anche in mobilità
